ABSTRACT
Premature ejaculation, also referred to as rapid or early ejaculation, is a poorly understood disorder with no single, widely-recognised, evidence-based definition. Studies based on patient self-reporting indicate that premature ejaculation is a common complaint with estimated prevalence ranging from 4%-39% of men in the general community.(1) However, a lack of an accurate validated definition has made comparison of the results of such studies difficult.(2) In addition, perception of normal ejaculatory latency varies by country and differs when assessed by the patient or their partner.(3) ▾Dapoxetine (Priligy-A. Menarini Farmaceutica Internazionale SRL), a short-acting selective serotonin reuptake inhibitor (SSRI) is the first drug to be licensed in the UK for on-demand management of diagnosed premature ejaculation.(4) In this article we review the evidence for dapoxetine and discuss some of the challenges associated with its introduction.

CLINICAL EVIDENCE

Dapoxetine
Although SSRIs are intended for chronic use in the treatment of depression and are designed to have pharmacokinetic profiles that would allow them to provide constant systemic concentration with long-term administration, they may need days to weeks to reach a maximum steady-state concentration in order to exhibit efficacy (Hiemke and Hartter 2000). Therefore, SSRIs are commonly used in a daily dosing schedule for the treatment of PE (Montague et al 2004). In addition to the potentially desirable side-effect of delaying ejaculation, this dosing regimen for long-acting SSRIs is associated with a number of undesirable side-effects, such as decreased libido and ED (Montague et al 2004). With the success of daily SSRI use in delaying ejaculation, it has been suggested that on-demand treatment with SSRIs possessing a short half-life and short Tmax would be equally effective, more convenient, and exhibit fewer serotonergic side-effects than observed with daily treatment (Waldinger 2005). Therefore, an ideal compound for the treatment of PE should exhibit pharmacokinetic profiles having rapid absorption, adequate availability to establish therapeutic exposure at the target site, and rapid elimination to reduce total drug exposure and minimize the incidence of side-effects (Modi et al 2006).
Dapoxetine, ((+)-(S)-N,N-dimethyl-(a)-[2-(1-naphthalenyloxy)ethyl]-benzenemethanamine hydrochloride) is a novel potent serotonin transport inhibitor with a unique pharmacokinetic profile, and is currently in development for use in the treatment of PE (Dresser et al 2004; Gengo et al 2005). Dapoxetine hydrochloride is a water-soluble powder with a molecular weight of 341.88 and has a pKa of 8.6 and is mainly charged at a physiological pH. These pharmacological characteristics allow for rapid distribution in the body and are different from the pharmacokinetics observed in conventional SSRIs. Dapoxetine is a fast-acting compound that attains its peak plasma concentration in about 1.5 hours after dosing, which is much faster than fluoxetine, paroxetine, or sertraline (Strassberg et al 1999, 2005; Eli Lilly 2005; GlaxoSmithKline 2005; Pfizer 2005). After oral administration, dapoxetine is readily absorbed, followed by a rapid decline in plasma concentrations. These pharmacokinetic properties make dapoxetine an excellent candidate for on-demand dosing, which avoids the potential disadvantages of continuous pharmacotherapy. The pharmacokinetic characteristics of dapoxetine compared with paroxetine and sertraline are provided in Table 2.

Although the pharmacological action of dapoxetine is similar to that of clomipramine and conventional SSRIs, the chemical structure and pharmacokinetic profile of dapoxetine differentiates it from other SSRIs. Dapoxetine is not a halogenated compound, whereas others contain one or more halogen atoms. The molecular structure of dapoxetine also includes a naphthyl moiety and it is possible that these features underpin the physicochemical and pharmacokinetic properties of the molecule.
Dapoxetine is a potent inhibitor of the 5-HT reuptake transporter. Studies conducted in cells expressing the human transcript of the 5-HT, norepineprine, and dopamine reuptake reporters to determine the inhibition constant (Ki) and the activity of dapoxetine (IC50) values showed that dapoxetine competed in a concentration-dependent manner for specific [3H]citalopram binding sites on the 5-HT reuptake transporter (Ki = 9.5), specific [3H]nisoxatine binding sites on the norepinephrine reuptake transporter (Ki = 6.6), and specific [3H]WIN35428 binding sites on the dopamine reuptake transporter (Ki = 5.8) (Gengo et al 2005). Additionally, dapoxetine inhibited the uptake of [3H]5-HT by the 5-HT reuptake transporter with a value of 1.12 nM, whereas the uptake of [3H]norepinephrine into cells utilizing the norepinephrine reuptake transporter and uptake of [3H]dopamine by the dopamine reuptake transporter were inhibited by dapoxetine with IC50 values of 202 nM and 1720 nM, respectively. In conclusion, dapoxetine binds to 5-HT, norepinephrine, and dopamine reuptake transporters and inhibits 5-HT, norepinephrine, and dopamine uptake with an order of potency: 5-HT > norepinephrine dopamine.
A double-blind, randomized, placebo-controlled, crossover, 3-period phase II US study of dapoxetine for the treatment of PE, comparing the safety and efficacy of on-demand dapoxetine 60 mg versus 100 mg versus placebo was presented (Hellstrom 2004). Each of the three 2-week treatment periods was separated by a 72-hour washout period. The patients enrolled in the study were all heterosexual men aged 18–65 years who had been involved in a stable, monogamous relationship for at least 6 months and whose baseline IELT was under 2 minutes, as measured by the partner using a stopwatch. The primary endpoint was IELT, as well as patients’ satisfaction control over ejaculation. A total of 130 men with PE completed the study. The mean baseline IELT increased from 1.01 minutes to 2.94 minutes with 60 mg dapoxetine, 3.20 minutes with 100 mg dapoxetine, and 2.05 minutes with placebo (p < 0.0001 vs placebo). Patients who received the 60-mg dose of dapoxetine reported significant increases in their control over ejaculation from baseline (p < 0.0001) and significant benefits in their sexual satisfaction (with their sexual intercourse episodes). Nausea, the most common reported adverse effect, occurred in 5.6% of patients who received 60 mg, 16.1% of those who received 100 mg, and 0.7% of those who received placebo (Table 3) (Hellstrom 2004). Of the 10 patients who discontinued the study because of side-effects, 9 were receiving the 100-mg dose of dapoxetine and 1 patient was receiving placebo.

Another two phase II clinical trials investigated the efficacy and tolerability of 20, 40, 60, and 100 mg dapoxetine in order to determine the appropriate on-demand doses of dapoxetine for further study in large-scale phase III clinical trials (Hellstrom et al 2005). Both double-blind, multi-center, randomized, placebo-controlled, 3-period cross-over phase II studies enrolled men with PE, based on DSM-IV-TR criteria and IELT. Subjects were instructed to take study drug prior to the anticipated sexual intercourse (1–3 hours prior in study 1, and 1–2 hours prior in study 2) and to attempt sexual intercourse ≥ twice weekly. The primary endpoint was IELT measured with a stopwatch held by the female partner. In study 1, 128 of 157 patients (20 mg and 40 mg dapoxetine), and in study 2, 130 of 166 (60 mg and 100 mg dapoxetine) randomized subjects completed the trials. All four doses of dapoxetine documented statistically significant improvements in IELT over placebo (Table 4).

In a recent study by Pryor et al (2005), the efficacy and tolerability of dapoxetine in the treatment of PE were evaluated. Men with PE (n = 2614) as defined by DSM-IV-TR criteria were enrolled in 2 randomized, double-blind, placebo-controlled, multi-center, phase III clinical trials of identical design, and each had a 2-week baseline period and 12-week treatment period. At the end of the baseline period, subjects with mean IELT ≥ 2 minutes were randomized 1:1:1 to placebo, dapoxetine 30 mg, or 60 mg, and instructed to take the study drug 1–3 hours prior to anticipated sexual intercourse. The primary efficacy endpoint was IELT adjusted for baseline, as measured by a stopwatch held by the female partner. Secondary endpoints were subjective perception of Control Over Ejaculation and Satisfaction With Sexual Intercourse, and were assessed at baseline, 4, 8, and 12 weeks, and rated on 5-point scales of between 0 (very poor) and 4 (very good). Preliminary results of this study demonstrated that there were significant differences between placebo and each active treatment group in mean IELT, Control Over Ejaculation, and Satisfaction With Sexual Intercourse (p < 0.0001). Changes from baseline to study end point for mean IELT were 0.90–1.75 min (placebo), 0.92–2.78 minutes (30 mg dapoxetine), and 0.91–3.32 minutes (60 mg dapoxetine). Both dapoxetine 30 mg and 60 were found to be more effective than placebo in increasing IELT on first dose. Treatment-related side-effects (incidence ≥5%) with both 30 mg and 60 mg dapoxetine were nausea (8.7% and 20.1%), headache (5.9% and 6.8%), diarrhea (6.8% with 60 mg), and dizziness (6.2% with 60 mg). Discontinuation rates due to adverse effect were reported as 0.0%, 4%, and 10% for placebo, 30 mg, and 60 mg doses of dapoxetine, respectively. The authors concluded that dapoxetine 30 mg or 60 mg is effective for the on-demand treatment of PE, with increased IELT, improved Control Over Ejaculation, and increased Satisfaction With Sexual Intercourse.
The use of ethanol and PDE-5 inhibitors with sexual activity is recognized. The interactions of dapoxetine with PDE-5 inhibitors and ethanol have also been investigated. The effects of PDE-5 inhibitors on the pharmacokinetics of dapoxetine were studied in a randomized, open-label, 3-period crossover study in healthy male subjects (n = 24; 18–45 years of age) receiving a single 60-mg dose of dapoxetine, dapoxetine 60 mg plus sildenafil 100 mg, and dapoxetine 60 mg plus tadalafil 20 mg (Dresser et al 2006). Each treatment was separated by a 6- to 14-day washout period. Blood samples were collected predose and at specified times after dosing for measurement of dapoxetine and its metabolites and tadalafil and sildenafil plasma concentrations. This study demonstrated that sildenafil and tadalafil did not have a clinically significant effect on the pharmacokinetics of dapoxetine. In the other study that was designed as double-blind, randomized, and 4-period crossover in healthy male adult subjects, the effect of ethanol on the pharmacokinetics of dapoxetine was investigated (Modi et al 2005). Each separate treatment period of a single dose of dapoxetine 60 mg and a single dose of 0.5 g/kg ethanol, dapoxetine 60 mg and ginger ale (placebo for ethanol), placebo (for dapoxetine) and 0.5 g/kg ethanol, and placebo for both drugs was administered with a 5- to 21-day washout period. Safety was assessed by adverse event reporting throughout the study and by laboratory tests, and blood samples were collected predose and at specified times after for measurement of plasma concentrations of dapoxetine and its metabolites and ethanol analysis. The results of this study demonstrated that co-administration of ethanol with dapoxetine did not produce significant changes in dapoxetine pharmacokinetics.

Conclusions
Neurobiological studies in animals and humans have provided better understanding of the physiology of ejaculation and pathophysiology of PE. The nature of PE is such that patients should not have to chronically take potent psychoactive drugs, with long-lasting actions and debilitating side-ef-fects. Dapoxetine is a selective serotonin transport inhibitor with unique pharmacokinetic and pharmocodynamic properties that account for its clinical effects. Future studies may demonstrate whether dapoxetine could possess many of the attributes of an ideal compound for the treatment of PE.
 
Psychologically it can be very frustrating as a man when you can't last long. Some women have a chemistry that just makes it difficult for me to last. I find it interesting how with some women I can last as long as I want, but with others I just get too turned on.

Well, I gave my research rat oxytocin last night and he had vivid sex dreams and wacked his little rat dick off twice. I'd like to give him dapoxetine and tadalafil as I notice his rat ding ding is hard quite often from tadalafil and most impressive sizewise considering he is just a kangaroo rat. I would imagine the relaxation from oxytocin would add to the length of time he could last but make sure he is with a special female rat as it makes him empathetic and loving....no rat bootie calls!!!
 
All SSRI's will allow you to be in complete control over time. SSRI's are usually given out for depression and/or anxiety. This is given out solely for premature ejaculation due to it's very short active life. If you suffer from PE this could genuinely be incredible for you and that is no BS.

INTRODUCTION: Men with comorbid erectile dysfunction (ED) and premature ejaculation (PE) may be concomitantly prescribed a phosphodiesterase type 5 (PDE5) inhibitor and dapoxetine.

AIM: Evaluate efficacy and safety of dapoxetine 30 mg and 60 mg on demand (prn) in men with PE and ED who were being treated with PDE5 inhibitors.

METHODS: This randomized, double-blind, placebo-controlled, flexible-dose, multicenter study enrolled men ≥18 years who met diagnostic criteria for PE including intravaginal ejaculatory latency time (IELT) of ≤2 minutes in ≥75% of sexual intercourse episodes; were on stable regimen of a PDE5 inhibitor; and had International Index of Erectile Function-erectile function domain score ≥21. Subjects received placebo, dapoxetine 30 mg, or dapoxetine 60 mg prn (1-3 hours before intercourse) for 12 weeks.

MAIN OUTCOME MEASURE: Stopwatch-measured average IELT, Clinical Global Impression of Change (CGIC) in PE, Premature Ejaculation Profile (PEP), and treatment-emergent adverse events (TEAEs).

RESULTS: Of 495 subjects randomized, 429 completed the study. Arithmetic mean average IELT significantly increased with dapoxetine vs. placebo at end point (5.2 vs. 3.4 minutes) and weeks 4, 8, and 12 (P ≤ 0.002 for all). Men who described their PE at least "better" using the CGIC were significantly greater with dapoxetine vs. placebo at end point (56.5% vs. 35.4%) and weeks 4, 8, and 12 (P ≤ 0.001 for all). Significantly better outcomes were also reported with dapoxetine vs. placebo on PEP measures. Incidence of TEAEs was 20.0% and 29.6% in placebo- and dapoxetine-treated subjects, respectively (P = 0.0135). TEAEs led to discontinuation in 1.6% of subjects in both groups. Most frequent TEAEs were known adverse drug reactions of dapoxetine treatment including nausea (9.2%), headache (4.4%), diarrhea (3.6%), dizziness (2.4%), and dizziness postural (2.4%).

CONCLUSIONS: In men with PE and comorbid ED on a stable regimen of PDE5 inhibitor, dapoxetine provided meaningful treatment benefit and was generally well tolerated.

Abstract
BACKGROUND: No drugs are approved for treatment of premature ejaculation. Our aim was to determine the efficacy and tolerability of on-demand dapoxetine in patients with severe premature ejaculation.

METHODS: We determined the efficacy of dapoxetine in a prospectively predefined integrated analysis of two 12-week randomised, double-blind, placebo-controlled, phase III trials of identical design done independently, in parallel, at 121 sites in the USA. Men with moderate-to-severe premature ejaculation in stable, heterosexual relationships took placebo (n=870), 30 mg dapoxetine (874), or 60 mg dapoxetine (870) on-demand (as needed, 1-3 h before anticipated sexual activity). The primary endpoint was intravaginal ejaculatory latency time (IELT) measured by stopwatch. Safety and tolerability were assessed. All analyses were done on an intention-to-treat basis. The trials are registered at ClinicalTrials.gov, numbers NCT00211107 and NCT00211094.

FINDINGS: 672, 676, and 610 patients completed in the placebo, 30 mg dapoxetine, and 60 mg dapoxetine groups, respectively. Dapoxetine significantly prolonged IELT (p<0.0001, all doses vs placebo). Mean IELT at baseline was 0.90 (SD 0.47) minute, 0.92 (0.50) minute, and 0.91 (0.48) minute, and at study endpoint (week 12 or final visit) was 1.75 (2.21) minutes for placebo, 2.78 (3.48) minutes for 30 mg dapoxetine, and 3.32 (3.68) minutes for 60 mg dapoxetine. Both dapoxetine doses were effective on the first dose. Common adverse events (30 mg and 60 mg dapoxetine, respectively) were nausea (8.7%, 20.1%), diarrhoea (3.9%, 6.8%), headache (5.9%, 6.8%), and dizziness (3.0%, 6.2%).

INTERPRETATION: On-demand dapoxetine is an effective and generally well tolerated treatment for men with moderate-to-severe premature ejaculation.
 
What is the dosage of this? Out of curiosity.

Inter-Vaginal Latency Time

For men with an average baseline IELT of 0.9 minutes, the average IELT improved to 3.1 minutes for a 30mg dose and 3.6 minutes for a 60 mg dose. Patients taking the placebo improved to 1.9 minutes.

30mg to 60mg. Start with 30mg.

Definitely start at 30mg. It may hit you hard at first (I have heard) so start off low and build up. I have heard a few guys literally say it has transformed them (confidence etc) so curious about guys trying out our dapoxetine.
 
This is honestly tempting to try, as well as the oxytocin. Funny that I'm on an AAS forum and I'm asking about these chemicals. Lol. I see that the dapoxetine is taken orally, how about the oxytocin. I appreciate the quick reply back on my question guys.

The oxytocin comes in powder form and is usually reconstituted with bacteriostatic water and injected subcutaneously. You can take oxytocin via nasal spray. You can reconstitute it and put it in a nasal spray bottle.
 
This study show it's safe to stack dapoxetine with tadalafil or sildenafil.

Potential pharmacokinetic interactions between dapoxetine, a serotonin transporter inhibitor developed for the treatment of premature ejaculation (PE), and the phosphodiesterase-5 inhibitors tadalafil and sildenafil, agents used in the treatment of erectile dysfunction (ED), were investigated in an open-label, randomized, crossover study (n=24 men) comparing dapoxetine 60 mg, dapoxetine 60 mg+tadalafil 20 mg, and dapoxetine 60 mg+sildenafil 100 mg. Plasma concentrations of dapoxetine, tadalafil, and sildenafil were determined by liquid chromatography-tandem mass spectrometry. Tadalafil did not affect the pharmacokinetics of dapoxetine, whereas sildenafil increased the dapoxetine AUCinf by 22%; these effects were deemed not clinically important. Dapoxetine did not appear to affect the pharmacokinetics of tadalafil or sildenafil. Most adverse events were mild in nature. Thus, dapoxetine has no clinically important pharmacokinetic interactions with tadalafil or sildenafil, and the combinations are well tolerated.

I told my mate about this and he wants it bad. He always says when he gets a new girl it is over very quick.

I take a small dose of citalopram for anxiety. It's probably not even needed now but I like it. Plus it causes no bad side effects and I can't be bothered with the withdrawal. That's an SSRI like Dapoxetine and let me tell you I literally have full control. I could go on for 3 hours straight if I wanted to. The big difference though between the two is dapoxetine is absorbed and eliminated rapidly in the body. Therefore it's not suitable for anxiety etc but used solely for premature ejaculation. I imagine for some PE could ruin their confidence and sex life so this really is a miracle drug and could change lives. Not even exaggerating there as we all know how important sex is and if you feel insecure about sex it must have a massive effect on confidence, dating and general happiness.

Dapoxetine lengthened normal ejaculation time by about 300% at a dose of 30mg taken one hour before intercourse, and about 400% at a dose of 60mg.

Premature ejaculation (PE) is considered to be the most common sexual problem affecting men, despite the likelihood that it is under-diagnosed. It is a complex condition with many physical and psychological components, making management complicated. It is important to develop treatments for PE as it adversely affects quality of life for individuals and partners. Dapoxetine is a short-acting selective serotonin reuptake inhibitor (SSRI) that has been developed principally for the treatment of PE. It is considered more suitable for the treatment of PE than other SSRIs as it can be used as an 'on demand' treatment to be taken a few hours before an expected sexual encounter, reducing the possibility of adverse effects. Dapoxetine may represent a breakthrough in the treatment of PE as it is the first drug to be licensed for this indication. This review attempts to present a balanced benefit-risk assessment of dapoxetine by examining the evidence from phase III clinical trials, focusing on its efficacy in prolonging intravaginal ejaculatory latency time (IELT), patient sexual satisfaction and safety in patients with PE. The benefits and risks of other therapies that are used to treat PE off-licence are also reviewed. There has only been one study to date that directly compares dapoxetine to another therapy, paroxetine, for this indication. It was found that dapoxetine is most effective at a dose of 60 mg in increasing IELT compared with placebo. All studies have also found that dapoxetine is well tolerated as an 'on-demand' therapy and with continual dosing; however, there are little data regarding possible long-term adverse effects. Findings of the dapoxetine development programme demonstrated that dapoxetine is associated with vasovagal-mediated (neurocardiogenic) syncope. No other associated significant cardiovascular adverse events were identified. Further research is needed to directly compare dapoxetine with other therapies and to investigate the outcomes of dapoxetine used in conjunction with behavioural therapies, and other non-pharmaceutical therapies.

Dapoxetine is a short-acting SSRI that can be taken as needed (prn) by men with PE. It has been studied in five separate multicenter, randomized, double-blind, placebo-controlled trials involving more than 6000 men with PE. In four studies that evaluated IELT as an endpoint (N = 4843), dapoxetine 30 and 60 mg prn achieved statistically significant increases in IELT versus placebo. Dapoxetine also showed statistically significant improvements in perceived control over ejaculation, PE-related personal distress, and other patient-reported outcomes in all five trials
 
Studies show it's safe to take dapoxetine with a pde5. That's the way I'd run it. Tadalafil is my favorite pde5 of the two I've tried. I still haven't had a chance to try the fastest acting and newest pde5, Avanafil.

Vicodin/percs, any of the opiate pain killers can have that effect. Keep in mind most women don't want to get pounded on for 40 min and you get frustrated wanting to nut and it ain't happening. Some women get worried it's them and their attraction/performance if you don't nut within a reasonable time frame.

Also, opiates would be a bad idea for regular ejac control. For every action there is an opposite and equal reaction. Taking it regularly then not using it will cause hypersensitivity leading to PE.

This was my point. Taking such a thing to stop you from cumming is OTT to me and not good at all. I don't like taking any strong painkillers. Sure if the guy used it once a month it's not ideal but not so bad. But if done regularly I can see major issues arising from that.

INTRODUCTION: With increasing interest and clinical research in male sexual disorders, it has become clear that not only psychological but also organic, neurobiological, and genetic factors may play an important role in premature ejaculation (PE).

AIM: This article provides an overview of the different treatment options both for lifelong (primary, "congenital") and acquired (secondary) PE.

METHODS: Review of the literature.

MAIN OUTCOME MEASURES: Currently used treatment options for PE.

RESULTS: Treatments reviewed include psychological/behavioral/sexual counseling therapy, topical anesthetics, dapoxetine, and other selective serotonin reuptake inhibitors (SSRIs), tricyclic antidepressants, and phosphodiesterase-5 (PDE-5) inhibitors.

CONCLUSIONS: Before starting any therapy for PE, correct diagnosis has to be made considering the patient's reported intravaginal ejaculatory latency time (IELT) and the duration and type of PE. Concomitant erectile dysfunction (ED) should be either ruled out or proven by appropriate means. In uncomplicated cases of PE with stable partnerships, medical treatment represents the first-choice option with a high likelihood of success. Dapoxetine, where available, or other SSRIs provide suitable therapeutic options with a good risk/benefit profile for patients. In complicated ("difficult-to-treat") PE patients, a combination of medication and sexual counseling should be considered the first treatment option. Combination therapies of PDE-5 inhibitors and PE-related medications should be offered to patients suffering from comorbid PE and ED, with ED treatment starting first. In those patients with severe PE-IELTs of <30-60 seconds or anteportal ejaculation-combination therapy of topical and oral medications can be offered and may considerably increase IELT, compared with either monotherapy.
 
Dapoxetine is an SSRI(antidepressant) with a half life of around two hours, whereas Prozac is an SSRI with a half life of around two weeks. The short half life explains why dapoxetine works for premature ejaculation as too long a half life can cause erectile issues.

Used it last night and all I can say it wow. The stuff definitely works. Made me last a hell of a lot longer than I thought it would, which helped with confidence. Went at it a couple times last night and a couple times this morning. All I did was take one dose of 30mg then 5mg about 5 hours later, don't know why. "just tried some dapoxetine for first time last night. It's a research chem that is being tested as a treatment for PE and it's been tested mostly on guys who can't last longer than 2 minutes before losing it. I've never really had an issue with not lasting long enough but I've had to use stop-start sometimes to keep it together which sometimes isn't the best.

I took a 60 mg dose at 8:30 last night and around 10:00 got started. We spent a good 20 minutes with her grinding on top however she wanted and I had zero urge to blast. She ended up with a charlie horse in her hamstring mid small orgasm and had to stop and recuperate and she thought that was the end of it.

We started again about 30 minutes later and once again, about 20 minutes of her riding and grinding to the hilt, about a 5 minute oral session, then another 10 minutes or so back to the riding until she finally collapsed in a ball drenching heap. I appreciate your review. I'll definitely have to get this when I get another girlfriend. I don't like casual sex and only sleep with a girl who I'm interested in having a relationship with. Dapoxetine (60 mg) 1-3 h before planned intercourse is a very effective treatment modality for PE. A guy in my dapoxetine thread at another forum posted that superior dapoxetine is the first he has used that was legit. He tried dapoxetine from two other research companies and they were both bunk. His only complaint was the bad taste. Hahaha the research liquids are diluted in grain alcohol for maximum absorption as the alcohol pulls the research chem directly into the blood stream via sublingual use. Unfortunately, grain alcohol tastes like burnt ass. Hahahaha :)

IMPORTANCE OF THE FIELD: Premature ejaculation (PE) is a common male sexual disorder which is associated with substantial personal and interpersonal negative psychological factors. Drug treatment of PE with an off-label antidepressant selective serotonin reuptake inhibitor (SSRI) drug is common. The lack of an approved drug and total reliance on off-label treatment represents a substantial unmet treatment need.

AREAS COVERED IN THIS REVIEW: Medline and the proceedings of major international and regional scientific meetings during the period 1994-2010 were searched for publications or abstracts using the word 'dapoxetine' in the title, abstract or keywords. This search was then manually cross-referenced for all papers. This review encompasses studies of dapoxetine pharmacokinetics, animal studies, human phase I, II and III efficacy and safety studies and drug-interaction studies. Dapoxetine is a potent SSRI, which is administered on demand 1-3 h before planned sexual contact. Dapoxetine is rapidly absorbed and eliminated, resulting in minimal accumulation and has dose-proportional pharmacokinetics, which are unaffected by multiple dosing. Dapoxetine 30 and 60 mg has been evaluated in five randomized, double-blind, placebo-controlled studies in 6,081 men aged > or = 18 years. Outcome measures included stopwatch-measured intravaginal ejaculatory latency time (IELT), Premature Ejaculation Profile (PEP) items, clinical global impression of change (CGIC) in PE, and adverse events. Mean IELT, all PEP items and CGIC improved significantly with both doses of dapoxetine versus placebo (p < 0.001 for all). The most common adverse events included nausea, dizziness and headache, and evaluation of validated rated scales demonstrated no SSRI class-related effects with dapoxetine use.

My mate keeps asking me for the dapoxetine so I just ordered some for him. He knows nothing about pep tides or research chems etc. I said to him try tadalafil with it so he is just going on my recommendation. He has no idea... I think he will be pleased :D

I ordered some dapoxetine from India (packaged as Prejac), along with an order of Super P-Force (a combination of sildenafil citrate (better known as Viagra) and dapoxetine). The Prejac pills were 60mg dapoxetine. The Super P was 100mg Viagra + 30mg dapoxetine. Honestly, when I tried the 60mg I was underwhelmed. Granted, we had intense foreplay followed immediately by her insisted hard pounding right away upon entry (when she's flying, it's almost impossible for me to control anything - she won't even let me back off to cool down a bit - when she's in the zone, she wants it HARD/NOW!), but I was looking for the ability to survive that first wave and move on to at least another... or more. When I took the Super P, I actually split the pill in half as I've read horror stories of woodies that won't go away - I don't like the idea of having to explain to the wife (and kids) why I can't get out of bed and go to work the next day -- or why I can't get up from my desk to meet with clients if I happen to spike one just because I had a fleeting thought of hammer-time with the wife... I noticed on another related forum one poster commenting on the putrid taste. Would it work to mix with OJ or something? I haven't taken anything in liquid form since I was a kid and loved grape flavored dimetapp. While I wouldn't want to do anything to reduce the potency of the liquid dapox, I also wouldn't want to try to explain to the Mrs. why she just "doesn't want to go there" if she swoops in for a kiss. I've learned that can happen from time to time when the bed is rockin'.

One last question - I began taking St. John's Wort (900 mg - 3x daily) about a month ago. My idea was to stack it with 30mg of dapox (and increasing to 60mg if I didn't end up frying my brain), hoping it would provide enough boost to get me the control I'm looking for. I know there are warnings against mixing St. John's Wort and other anti-depressants, but with the minimal half-life of dapoxetine, I figured it may not be as big of a deal. Thoughts?
 
I'm not sure on the St. John's wart. But with dapoxetine you can put it in juice and drink it. It tastes like cheap dirty vodka. It would taste like a mixed drink hahahaha :). The absorption of liquid orals is sped up and enhanced by holding the liquid under the tongue for 30 seconds but drinking it works too.
