The big problem that most people face is a lack of proper equipment. You just can't cook up some test in your kitchen and expect it to turn out. Even if you have the necessary equipment or like me, have access to a properly equipped lab, there's little chance of it being sterile enough to shoot. I believe the foremost authority on this topic is Patrick Arnold. Maybe you should post your question to him and ask why. I'm sure he get's bombarded with questions all the time, so I'm not sure if he will answer yours or not.
