Every new diet drug that comes out is supposedly the next big thing. However, two new drugs have the potential to shake up the dieting industry. They are Pramlintide and Metreleptin and it appears they can help people drop more pounds than ever before. And the best part is that they’ll work on burning fat without you having to lift a finger!
Lifting, eating, and bulking are some of the best things about bodybuilding. Watching my muscles grow and being able to eat massive amounts of calories are the main reasons why I love bodybuilding so much. But for every up, there’s a down. And that down for me in bodybuilding is cutting.
I mean who actually likes to cut their calories while turning up the cardio? Sure it’s great to see the end results of cutting calories and doing more cardio, but it is hell getting there. That is why I like to monitor what’s going on in the diet industry so I can see if any useful drugs will be coming out. Yes, that’s right... I can be one of those lazy guys who would rather burn fat through diet drugs rather than doing an extra hour of cardio in the gym - expecially at this time of year.
And while I’ve been watching the diet industry, I recently saw where there are some new drugs which can cause massive weight loss in people - Pramlintide and Metreleptin. Weight loss expert Dr. Ken Fujoka stated this fact when he said, “With these new medications, now we're getting what we call double digit weight loss. 10 percent and up.” Fujoka continued by saying, “So now you're losing 20, 30, and 40 pounds of weight with these new weight loss medications, which really makes the doctor and the patient a lot happier.”
The Pramlintide and Metreleptin combo drug that is allowing people to see these double digit losses in weight is made by a company called Amylin. Their injectable drug doesn’t have a name yet, but it has proven to curb people’s appetites in clinical trials. The injectable is a combination of two drugs Pramlintide, a synthetic analog of amylin, a neurohormone that regulates appetite and food intake, and Metreleptin a recombinant form of human leptin. By using Amylin’s drug, people can resist their urges and start cutting major weight through their decreased appetite. The CEO of Amylin, Daniel Bradbury, explained the drug’s effects by saying, “These are molecules, which actually act like a naturally occurring molecule (leptin). This is a molecule your fat cells produce to control your body weight.”

Pramlintide / Metreleptin
The pramlintide/metreleptin combination reflects the tremendous potential of Amylin’s integrated neurohormonal strategy in developing novel, safe, and effective new therapies for weight loss.
- Pramlintide, the active ingredient in SYMLIN®, is a synthetic analog of amylin, a neurohormone secreted by the pancreas that is known to play a key role in the regulation of appetite, food intake and postprandial glucose concentration.
- Metreleptin is an analog of human leptin, a neurohormone secreted by fat cells that plays a fundamental role in the regulation of energy homeostasis, fat and glucose metabolism and body weight.
In a proof-of-concept study completed in late 2008, patients treated with a combination of pramlintide and metreleptin reduced body weight on average by 12.7 percent over 24 weeks—representing an average loss of 25 pounds. Weight loss continued through the end of the study, with no evidence of plateau.
Based on these significant results, a 28-week Phase 2B pramlintide/metreleptin clinical study was completed in late 2009, followed by an extension study to 52 weeks, in which the pramlintide/metreleptin combination met the key target criteria of sustained and robust weight loss.
Patients who continued treatment with pramlintide/metreleptin for a total of 52 weeks demonstrated sustained weight loss,whereas those continuing on placebo regained almost all of their weight. Consistent with results at 28 weeks, the most robust efficacy was seen in patients with a body mass index (BMI) less than 35 kg/m2.
The combination therapy appeared to be generally well tolerated through 52 weeks, with nausea and injection site adverse events observed as the most common side effects on initiation of pramlintide or metreleptin in combination treatment. Following initiation of therapy, these adverse events occurred at a reduced rate over time in patients continuing combination therapy.
In November 2009, Amylin entered into a worldwide agreement with Takeda to co-develop and commercialize pharmaceutical products for the treatment of obesity and related indications. The companies selected the pramlintide/metreleptin obesity program for advancement toward phase 3 clinical studies in early 2010, and enabling activities are underway.
In addition to the promising drug by Amylin, Arena Pharmaceuticals is also putting their company back in the news with the development of Lorcaserin. Lorcaserin is a traditional weight loss pill that is in the final stages of FDA trials, and is producing the aforementioned double digit weight loss results.
Dr. Dominic Behan gave a clear picture of how Lorcaserin works by explaining, “As a species we tend to preserve body fat and you need some way to short circuit that process, and Lorcaserin does that by modulating brain systems that can prevent food intake. So it decreases your appetite, so you take in less food.”
As mentioned before, the results of both Lorcaserin and Amylin’s drug are incredible with people in trials losing up to 10% of their body weight. But aside from making people look better, Daniel Bradbury pointed out that the aim of these drugs goes beyond mere cosmetic purposes. Bradbury said that these drugs are huge in reducing blood pressure, lowering cholesterol, and improving overall health status.
Personally, I just care about the looking better aspect, but I’d definitely take improved health along with the dramatic weight loss results offered by these drugs. All I know is that I’m excited to see these drugs hit the market – especially Lorcaserin since it is almost through the FDA trials!
